Definately excess iodine has been shown to cause abortion/ miscarriage. I have been working on iodine excess and both steroidogenesis and reproductive functions both in animal models and epidemiologic studies has been shown to be affected under iodine excess.

I am a third year graduate student in a lab that focuses on growth hormone. My project, however, is extremely thyroid hormone specific. From most of the literature I have found, it seems thyroid hormones are measured by RIA instead of ELISA. This leads me to two questions:
Is there a specific reason RIA is used over ELISA?
Can an ELISA be used to measure serum T4 and T3 levels in mice effectively?

I am now having a human serum with the mixture of T3 and T4 with T4 concentration is 100 times bigger than T3 concentration.
The anti-T3 antibody that I already had does not have high specificity, so I got interfered by T3 with results in bad results.

As I read in papers, before doing the radioimmunoassay (RIA) to detect thyroid hormones such as T3, we should free the hormones from binding the carrier proteins like thyroxine-binding protein (TBG) and albumin for higher accuracy. And 8-Anilino-1-naphthalenesulfonic acid (ANS) is one of the most widely used chemical reagents that can do that liberating purpose.
I searched several papers, however, they did not describe how ANS could do that such function?
Is it like a competitve substance that goes into protein and occupies T3 binding site? Or it will change the structure of the protein? Or something else...
If anyone has the explanation or documents/papers/books related to this issue, please share to me.

Dear Dr Furlan, your point of view about TSH resistance seems to be a strong recognition. However, I have to search more to find a reasonable mechanism which justify that how oxymorphone can causes TSH resistance!

Hello Andrew,
We have measured the release of T4 from static incubations of fish' thyroid tissue, and measuring total T4 concentrations in the incubation medium with a commercial ELISA. As Danielle says: validation is important. In this case we prepared standards in the same medium (matrix) as the final medium the samples were collected in.
I think this should work for superfusion experiments as well.

The choice of which method to choose for multiple comparisons depends upon you making a judgement as to which is best in your case, and upon some practical considerations, like which method is commonly used in your field.
I know. That's very helpful.
Bonferroni is very conservative.  That is, you are very unlikely to find false differences between groups.  But you are also likely to miss significant differences that you would find with another method.  In my opinion, it is too conservative in most cases.  But again, this is a decision you have to make for your own research.  If it is important to you to not find any false differences, then a conservative method is good for you.  However, if it is more important to not exclude differences that are probably real, then a less-conservative approach is better for you.
My best advice to read about the methods and choose the one you find would be best in your research.

Dear colleague,
According to DePaolo and Masoro 1989, the total T4 range in Swiss and C mice are 5.5±0.7 and 4.7±0.3 µg/ml, respectively. Total T3 level range from 65-85 ng/100 ml in both SW and ICR mice these measurements were applied by RIA Kit.
Of course, the strain and age can be change the reference ranges of T3 and T4. Moreover, many factors including auto-antibodies are known to interfere with thyroid hormone assays.

I think that there is definitely a relationship . In  hyperinsulinemic/diabetic patients  the epimerase enzyme activity increased . This implies an increase of the conversion of myo - inositol to D - chiro - inositol . This represents a defense mechanism to reduce insulin resistance . At the same time , however, the reduction of myo - inositol determines a reduction of myo - inositolphsphoglycan promoters of the signals for both FSH and TSH . This results in a resistance of the thyroid to TSH with consequent increase of the same . I hope to be liked . BR Vittorio

Since all researches available to me said that creatine monohydrate has no significant effect on triglycerides or T3 and one of these researches showed decreases in T3 and increases in T4, where the authors suggested the causes stands behind that results , yet I didn’t find a potential mechanism responsible for the increase of triglycerides and T3 in sera of broiler chicks at 42nd day and if there is an explain please just attach the references related to that.

I'm not sure if anyone has measured thyroid hormones in hair or urine - that would be interesting. However, I would consider the fact that thyroid hormones directly affect hair follicle function and urinary excretion patterns. Could make the analysis and interpretation very challenging and certainly complex. Good luck!

thanks for your quick reply (and sorry for my delayed one). I have already checked the list you mentioned beforehand, but there is no reference refering to an administration of thyroid hormones for a duration of 4 weeks with pump model 2006. The technical support at Charles River's was also uncertain..
My main concern is about the stability of the hormones in vivo: 
Is it necessary to replace the hormone solution during treatment, or not?

Hello, T4 is much better than T3 because thyroid gland release 90-95 % T4 and 5-10% T3. Hence, T4 is the best indicator of thyroid function. Too late, T4 is converted to T3 in the inner of the cells by deiodinase type II. Good Lucky.

In clinical practice, frequently supplementation with synthetic or natural thryroid hormone is used. This inevitably results in a low TSH because of feedback inhibition by triiodothyronine (T3) on synthesis and release of T4 from the thyroid gland. Endocrinologists often express a concern that "the TSH is too low", ie below the accepted clinical range. None have been able to explain to me what are the long term consequences of having a low TSH in and of itself. What is too low? Since it must be expected in thyroid supplementation, I fail to see the concern, and I cannot find relevant publications that would explain to me the ramification of low TSH. I call on the world-wide expertise of ResearchGate participants.

I would like to measure the concentration of T3 and T4 in zebrafish, and I'm trying to figure out the best way to extract the hormones from fish tissues. I tried extracting in phosphate buffer, but the concentrations were too low. Most of the protocols I find use barbital buffer, which I would like to avoid if at all possible, since it is a controlled substance and requires DEA certification. Any assistance would be greatly appreciated!

Hi Sarah,
the problem might be that most of the thyroid hormones are protein bound (serum albumin, receptors etc). So you'll need something that displaces the hormones from the proteins. Maybe high salt will do it, or adding chemically similar compounds which do not interfere with your assay.
Another thought was passing the tissue homogenate over immobilized antibodies, as they should have specific and probably stronger binding properties. Then elute the hormones with acidic buffer, then re-equilibrate to column to neutral for the next batch.
Edit: Get and compare the protocols for the determination of free and total thyroid hormones in serum from a department for lab diagnostics.
