Three novel species of Aquapteridospora (Distoseptisporales, Aquapteridosporaceae) from freshwater habitats in Tibetan Plateau, China

Abstract and Figures

During an investigation of lignicolous freshwater fungi in the Tibetan Plateau, three Aquapteridospora taxa were collected from freshwater habitats in Xizang, China. The new species possess polyblastic, sympodial, denticles conidiogenous cells and fusiform, septate, with or without sheath conidial, that fit within the generic concept of Aquapteridospora , and multi-gene phylogeny placed these species within Aquapteridospora . Detailed morphological observations clearly demarcate three of these from extant species and are hence described as new taxa. The multi-gene phylogeny of the combined LSU, TEF 1-α, and ITS sequence data to infer phylogenetic relationships and discuss phylogenetic affinities with morphologically similar species. Based on morphological characteristics and phylogenetic analyses, three new species viz. A. linzhiensis , A. yadongensis , and A. submersa are introduced. Details of asexual morphs are described, and justifications for establishing these new species are also provided in this study.

Aquapteridospora-
homycetidae genera incertae sedis, based on morphological and phylogenetic

Materials and methods
Collection, morphological examination and isolation
Submerged decaying wood samples were collected from freshwater habitats in
southeast Xizang, China. Fresh specimens were studied following the methods

The Tibetan Plateau is renowned for its distinctive biological diversity and

         

understudied in this region, primarily due to the limited number of researchers
focusing on freshwater fungi in the Tibetan Plateau. During our investigation
into freshwater fungal diversity on the Tibetan Plateau, we introduced three
new species within the genus Aquapteridospora, supported by both phylogenet-
ic analysis and morphology. The discovery of these new species revealed the

region are expected in the future.

Research into freshwater fungi has generated a wealth of information over the past decades with various published articles, i.e., reviews, books, and monographs. With the advancement of methodologies used in freshwater fungal research, and numerous mycologists working on this ecological group, our knowledge progress and understanding of freshwater fungi, including novel discoveries and new insights in the ecology of freshwater fungi, has advanced. With this enormous progress, it is timely that an updated account of freshwater fungi be compiled in one volume. Thus, this account is published to give a comprehensive overview of the different facets of freshwater fungal biology. It includes an updated classification scheme based on the latest taxonomic and phylogenetic analysis of freshwater fungal taxa, including their evolutionary history. The biology, diversity, and geographical distribution of higher and basal freshwater fungi are also discussed in the entries. A section on dispersal and adaptation of filamentous freshwater fungi is included in the present work. The ecological importance and role of fungi in the breakdown of wood in freshwater habitats, including their physiology, are discussed in detail. The biotechnological potential of freshwater fungi as producers of bioactive metabolites are reviewed, with methodologies in antimicrobial drug discovery. The present volume also provides an overview of different high throughput sequencing (HTS) platforms for freshwater fungal research highlighting their advantages and challenges, including recent studies of HTS in identification and quantification of fungal communities in freshwater habitats. The present volume also identifies the knowledge gaps and direction of future research in freshwater fungi.

Eight hyphomycetes were collected as part of an investigation into the diversity of hyphomycetous fungi in China. Based on morphology and multi-loci (LSU, ITS, tef1α, and rpb2) phylogenetic analyses, five new taxa, including a new Aquapteridospora species A. hyalina and four novel Distoseptispora species, viz D. aquisubtropica, D. septata, D. tropica, and D.wuzhishanensis were introduced in Distoseptisporales (Sordariomycetes). Two new habitat records, viz Distoseptispora pachyconidia and D. xishuangbannaensis were firstly reported. Also provided in this study are detailed descriptions of eight new collections and a revised phylogenetic tree for the Distoseptisporales.

During an investigation of freshwater fungi in Jiangxi province, China, a new hyphomycetous fungus, Aquapteridospora jiangxiensis, was collected and isolated. Aquapteridospora jiangxiensis is characterized by its unbranched and guttulate conidiophores with multi-septa swollen at the base, polyblastic conidiogenous cells with sympodial proliferations, and denticles, and guttulate conidia with a sheath. A photo plate of the macro- and micro-morphology and a muti-loci (ITS, LSU, SSU, TEF1 and RPB2) phylogenetic tree are provided. A key to the species of Aquapteridospora is also presented in this paper.

A comprehensive account of fungal classification from freshwater habitats is outlined and discussed in the present review based on literature of biodiversity studies and recent morpho-phylogenetic analyses. A total of 3,870 freshwater fungal species are listed with additional details on the isolation source, habitat, geographical distribution, and molecular data. The Ascomycota (2,968 species, 1,018 genera) dominated the freshwater fungal taxa wherein Sordariomycetes (823 species, 298 genera) had the largest number, followed by Dothideomycetes (677 species, 229 genera), Eurotiomycetes (276 species, 49 genera), and Leotiomycetes (260 species, 83 genera). Other phyla included in the updated classification of freshwater fungi are: Chytridiomycota (333 species, 97 genera), Rozellomycota (221 species, 105 genera), Basidiomycota (218 species, 100 genera), Blastocladiomycota (47 species, 10 genera), Monoblepharomycota (29 species, 6 genera), Mucoromycota (19 species, 10 genera), Aphelidiomycota (15 species, 3 genera), Entomophthoromycota (6 species, 4 genera), Mortierellomycota (5 species, 3 genera), Olpidiomycota (4 species, 1 genus), Zoopagomycota (3 species, 2 genera), and Sanchytriomycota (2 species, 2 genera). The freshwater fungi belong to 1,361 genera, 386 families and 145 orders. The Pleosporales and Laboulbeniaceae are the largest freshwater fungal order and family comprised of 391 and 185 species, respectively. The most speciose genera are Chitonomyces (87, Laboulbeniomycetes), Verrucaria (50, Eurotiomycetes), Rhizophydium (52, Rhizophydiomycetes), Penicillium (47, Eurotiomycetes), and Candida (42, Saccharomycetes).

This paper provides an updated classification of the Kingdom Fungi (including fossil fungi) and fungus-like taxa. Five-hundred and twenty-three (535) notes are provided for newly introduced taxa and for changes that have been made since the previous outline. In the discussion, the latest taxonomic changes in Basidiomycota are provided and the classification of Mycosphaerellales are broadly discussed. Genera listed in Mycosphaerellaceae have been confirmed by DNA sequence analyses, while doubtful genera (DNA sequences being unavailable but traditionally accommodated in Mycosphaerellaceae) are listed in the discussion. Problematic genera in Glomeromycota are also discussed based on phylogenetic results.

Distoseptispora is one of the sporidesmium-like taxa with great variation in asexual morphology and delineation of species. Phylogenetic analyses of four gene regions LSU, ITS, TEF1α, and RPB2 revealed the placement of several sporidesmium-like species in Distoseptispora (Distoseptisporaceae, Distoseptisporales, Sordariomycetes), collected on submerged decaying twigs from streams in China and Thailand. Based on morphological examination and molecular DNA data, six new species, Distoseptispora amniculi, D. atroviridis, D. effusa, D. fusiformis, D. hyalina, and D. verrucosa, are proposed. Among them, D. hyalina is the first sexual morph confirmed in the genus. A new geographical record is reported for D. lignicola in China. Conidial length proved to be of less taxonomic significance for some Distoseptispora species, whereas the type of conidial septa is informative at species level.

Freshwater fungi comprises a highly diverse group of organisms occurring in freshwater habitats throughout the world. During a survey of freshwater fungi on submerged wood in streams and lakes, a wide range of sexual and asexual species were collected mainly from karst regions in China and Thailand. Phylogenetic inferences using partial gene regions of LSU, ITS, SSU, TEF1α, and RPB2 sequences revealed that most of these fungi belonged to Dothideomycetes and Sordariomycetes and a few were related to Eurotiomycetes. Based on the morphology and multi-gene phylogeny, we introduce four new genera, viz. Aquabispora, Neocirrenalia, Ocellisimilis and Uvarisporella, and 47 new species, viz. Acrodictys chishuiensis, A. effusa, A. pyriformis, Actinocladium aquaticum, Annulatascus tratensis, Aquabispora setosa, Aqualignicola setosa, Aquimassariosphaeria vermiformis, Ceratosphaeria flava, Chaetosphaeria polygonalis, Conlarium muriforme, Digitodesmium chishuiense, Ellisembia aquirostrata, Fuscosporella atrobrunnea, Halobyssothecium aquifusiforme, H. caohaiense, Hongkongmyces aquisetosus, Kirschsteiniothelia dushanensis, Monilochaetes alsophilae, Mycoenterolobium macrosporum, Myrmecridium splendidum, Neohelicascus griseoflavus, Neohelicomyces denticulatus, Neohelicosporium fluviatile, Neokalmusia aquibrunnea, Neomassariosphaeria aquimucosa, Neomyrmecridium naviculare, Neospadicoides biseptata, Ocellisimilis clavata, Ophioceras thailandense, Paragaeumannomyces aquaticus, Phialoturbella aquilunata, Pleurohelicosporium hyalinum, Pseudodactylaria denticulata, P. longidenticulata, P. uniseptata, Pseudohalonectria aurantiaca, Rhamphoriopsis aquimicrospora, Setoseptoria bambusae, Shrungabeeja fluviatilis, Sporidesmium tratense, S. versicolor, Sporoschisma atroviride, Stanjehughesia aquatica, Thysanorea amniculi, Uvarisporella aquatica and Xylolentia aseptata, with an illustrated account, discussion of their taxonomic placement and comparison with morphological similar taxa. Seven new combinations are introduced, viz. Aquabispora grandispora (≡ Boerlagiomyces grandisporus), A. websteri (≡ Boerlagiomyces websteri), Ceratosphaeria suthepensis (≡ Pseudohalonectria suthepensis), Gamsomyces aquaticus (≡ Pseudobactrodesmium aquaticum), G. malabaricus (≡ Gangliostilbe malabarica), Neocirrenalia nigrospora (≡ Cirrenalia nigrospora), and Rhamphoriopsis glauca (≡ Chloridium glaucum). Ten new geographical records are reported in China and Thailand and nine species are first reported from freshwater habitats. Reference specimens are provided for Diplocladiella scalaroides and Neocirrenalia nigrospora (≡ Cirrenalia nigrospora). Systematic placement of the previously introduced genera Actinocladium, Aqualignicola, and Diplocladiella is first elucidated based on the reference specimens and new collections. Species recollected from China and Thailand are also described and illustrated. The overall trees of freshwater Dothideomycetes and Sordariomycetes collected in this study are provided respectively and genera or family/order trees are constructed for selected taxa.
