Abstract
Phosphorus solubilizing bacteria Pantoea agglomerans isolated from the onion rhizosphere
and were screened for antagonistic activity against Fusarium oxysporum f.sp. cepae(FOC).
The isolate inhibited mycelial growth of FOC at rates from 20% in in vitro conditions. The
bacterial strain Pantoea agglomerans were defined potential to solubilizing phosphate use of
NBRIP(National Botanical Research Institut . This izolate was
identified by using biochemical tests(levan production, oxidase test, catalase test and gram
test) and BIOLOG profiling system as Pantoea agglomerans. We have found the phosphorus
solubilization index 3.3 and 286.89 mg/l for the soluble phosphate solubilizing activity of
Pantoea agglomerans. In pot experiments bacterization inoculation of onion seeds
significantly increased the basal rot disease of onion plants by 60% according to the positive
control. These results demonstrated the potential for biological control using these P-
solubilizing rhizobacteria as inoculants for onion basal rot disease as commercial plant
activator ISR2000(Improcrop).
Keywords: Phosphorus solubilizing bacteria, Pantoea agglomerans, Fusarium oxysporum
f.sp. cepae
Introduction
Phosphorus (P) is one of the most main nutrients needed for the plant growth and
development(Illmer and Schinner, 1992). Root growth, stem force, seed formation, yield
maturity and resistance to plant diseases are the associated with phosphorus feeding.(Hao et
al., 2002). However, the applied soluble forms of P fertilizers are easily turn into insoluble
forms(Omar, 1997).Many species of microorganisms are known to live plant rhizosphere
and play significant role in growth via the P solubilization and nitrogen fixation
additionally produced different antagonistic substances against soil pathogen(Hussain et al.,
2016). Phosphate solubilizing bacteria (PSB) are used as biofertilizer. These PSBs release
different types of organic acids as oxalic acid, lactic acid, citric acid, gluconic acid etc. and
therefore PSBs is called plant growth promoting rhizobacteria (PGPR)(Zheng et al., 2019).
In recent years, these bacteria it was used biological control of plant pathogens besides the
effect of plant growth promoting (Ongena and Jacques, 2008). The use of PGPRs for the
plant growth and biological control of plant diseases is thought alternative to the use of
chemical fertilizers(Raaijmakers et al., 2009). The objective of this research was to evaluate
the biocontrol effect of phosphate solubilising strain Pantoea agglomerans against Fusarium
oxysporum f.sp.cepae (FOC) at the in vivo and in vitro conditions. This plant pathogen is a
serious pathogen, which causes basal rot on onion. The effect of Pantoea agglomerans against
of FOC was compared to the commercial plant activator ISR 2000 (Improcrop).

Onion rhizosphere soil solution when inoculated on the NBRIP medium was develop many
groups of bacteria. A strain, which was named IB-61, had a marked soluble phosphate
solubilizing activity as by the clear zone around the colony after 8 day incubation at 25 oC.
We have found the phosphorus solubilization index 3.3 by dividing the zone diameter to the
diameter of this bacteria(Figure 1). We also analyzed average 286.89 mg/l for the soluble

Various studies have been done investigating the importance of PSBs, its use as biofertilizer,
identi (Liu et al., 2014).
Our results conform to (Ranjan et al., 2013) who observed and also isolated PSBs from
various soil samples and selected 12 effective PSB isolates on their ability to produce a clear
quantitative estimation of P-solubilization ranged
Similar to our results, (Verma et al., 2018a)
showed that the PSBs also showed inhibition of the phytopathogens Fusarium oxysporum
and Rhizoctonia solani. Researchers observed that the highest antagonistic activity was
exhibited by an un identified strain TX-II (62.0%) against Fusarium oxysporum, whereas
Bacillus sp showed the highest inhibition (54.44%) of Rhizoctonia solani. More recently it
was found that the phosphate-solubilizing Pantoea agglomerans isolate was found to be the
most effective in inhibiting three fungal pathogens: Fusarium oxysporum, Curvularia sp. and
Alternaria sp.at a rate of 29.25, 54.9 and 40% respectively (Verma et al., 2018b).
Results and Suggestions
This biocontrol potential of PSBs might be due to stimulate the systemic resistance in plants
through increasing the phosphorus uptake from the soil by dissolving the insoluble
phosphorus in the soil. This is important in using PSBs as biofertilizers in agriculture to
reduce the application of chemical inorganic P and to decrease environmental pollution.

Background and Aims: Leersia oryzoides, a wild relative of rice (Oryza sativa), may carry potential seed-borne bacterial endophytes which could be used to enhance growth of rice. We hypothesized that seed-associated bacteria from L. oryzoides would be compatible with rice and promote seedling growth, development, and survival. Methods: We isolated bacteria from seed of L. oryzoides and checked compatibility with rice as well as Bermuda grass seeds for seedling growth promotion. Internal colonisation of bacteria into root cells was observed by ROS staining and microscopic observation. Growth promoting bacteria were evaluated for IAA production, phosphate solubilization and antifungal activities. Results: Overall, ten bacteria were found to be growth promoting in rice seedlings with effects including restoration of root gravitropic response, increased root and shoot growth, and stimulation of root hair formation. All bacteria were identified by 16S rDNA sequencing. Six bacteria were found to become intracellular in root parenchyma and root hairs in rice and in Bermuda grass seedlings. Six bacteria were able to produce IAA in LB broth with highest (47.06±1.99 µg ml-1) by LTE3 (Pantoea hericii). Nine isolates solubilized phosphate and inhibited at least one soil borne fungal pathogen. Conclusions: Seed bacteria of L. oryzoides are compatible with rice. Many of these bacteria become intracellular, induce root gravitropic response, increase root and shoot growth, and stimulate root hair formation in both rice and Bermuda grass seedlings. Presence of bacteria protects seedlings from soil pathogens during seedling establishment. This research suggests that bioprospecting microbes on near relatives of rice and other crop plants may be a viable strategy to obtain microbes to improve cultivation of crops.

Trichoderma sp., a well known biological control agent against several phytopathogens, was tested for its phosphate (P) solubilizing potential. Fourteen strains of Trichoderma sp. were isolated from the forest tree rhizospheres of pinus, deodar, bamboo, guava and oak on Trichoderma selective medium. The isolates were tested for their in-vitro P-solubilizing potential using National Botanical Research Institute Phosphate (NBRIP) broth containing tricalcium phosphate (TCP) as the sole P source, and compared with a standard culture of T. harzianum. All the cultures were found to solubilize TCP but with varying potential. The isolate DRT-1 showed maximum amount of soluble phosphate (404.07 µg.ml-1), followed by the standard culture of T. harzianum (386.42 µg.ml-1) after 96 h of incubation at 30+1(0)C. Extra-cellular acid and alkaline phosphatases of the fungus were induced only in the presence of insoluble phosphorus source (TCP). High extra-cellular alkaline phosphatase activity was recorded for the isolate DRT-1 (14.50 U.ml-1) followed by the standard culture (13.41 U.ml-1) at 72h. The cultures showed much lesser acid phosphatase activities. Under glasshouse conditions, Trichoderma sp. inoculation increased chickpea (Cicer arietinum) growth parameters including shoot length, root length, fresh and dry weight of shoot as well as roots, in P-deficient soil containing only bound phosphate (TCP). Shoot weight was increased by 23% and 33% by inoculation with the isolate DRT-1 in the soil amended with 100 and 200 mg TCP kg-1 soil, respectively, after 60 d of sowing. The study explores high P-solubilizing potential of Trichoderma sp., which can be exploited for the solubilization of fixed phosphates present in the soil, thereby enhancing soil fertility and plant growth.

Phosphate-solubilizing bacterial strains (PSB) were isolated from maize, onion, jasmine, and tomato rhizosphere soils from four different localities of Salem (Tamil Nadu) - (i) Ammapalayam, (ii) Attayampatty, (iii) Seeragapadi, and (iv) Ariyanoor. Twelve efficient PSB isolates were selected from the colonies based on their ability to form clear zone on Pikovskaya's agar medium. The isolated PSB released high amount of phosphorus from calcium phosphate. The efficient bacterial strains isolated from rhizosphere soils released high amount of phosphorus. Oligonucleotide primers in RAPD characterization showed genetic variation among the bacterial strains. Amplified fragments showed 40% shared and 20% unique polymorphic bands. The rest 40% was monomorphic bands.

Vermicomposting is a dependable waste recycling technology which greatly augments N and P levels mainly through microbial action. This paper aims to identify efficient N-fixing (NFB) and P-solubilizing (PSB) bacteria from earthworm intestines. Various combinations of vegetable market waste, rice straw, and cowdung were fed to two earthworm species (Eisenia fetida and Perionyx excavatus). Total organic C decreased, pH shifted towards neutrality, and NPK availability, and microbial (NFB, PSB, and total bacteria) population increased remarkably during vermicomposting with E. fetida. Therefore, 45 NFB and 34 PSB strains isolated from Eisenia gut were initially screened, their inter-dominance assessed, and 8 prolific strains were identified through 16SrRNA sequencing. Interestingly, two novel N-fixing strains of Kluyvera ascorbata emerged as an efficient biofertilizer candidate. Moreover, both N-fixing and P-solubilizing strains of Serratia and Bacillus were isolated from earthworm gut. All the isolated strains significantly improved soil health and facilitated crop growth as compared to commercial biofertilizers.

Phosphate-solubilizing bacteria (PSB) were isolated and characterized from the rhizosphere and bulk soils of Areca catechu plants. A long history of phosphate fertilizer use has elicited a direct effect on the incidence of soil PSB. Their abundance and ability to solubilize insoluble phosphate were significantly greater (P < 0.0001) in soils with low available phosphorus (P) content than in other soil types. Three efficient PSB strains, namely, ASL12, ASG34, and ADH302, were identified as Acinetobacter pittii, Escherichia coli, and Enterobacter cloacae by characterizing 16S rRNA sequences and biochemical characteristics; they produced gluconic acid at concentrations of 7862.4, 4306.5, and 2663.8 mg L-1, respectively. The highest amount of solubilized P was determined in Pikovskaya (PVK) medium for the bacterial strain ASL12. The secretion of gluconic acid was related to the available P of rhizosphere soils and P solubilization. Under shaded conditions, the application of these three strains significantly improved plant height, shoot and root dry weight, and nutrient uptake of A. catechu seedlings. A further increase in P solubilization was observed by co-inoculating the three strains and also applying tricalcium phosphate (TCP) or aluminum phosphate (AP). A significant (P < 0.05) correlation was also observed between P-solubilization activity and A. catechu plant growth in pot experiments. Thus, the three strains can be potentially applied as inoculants in tropical and aluminum-rich soils.

A total of 36 fungal species isolated from soil were tested for their ability to solubilize rock phosphate (RP) in agar plates. Most of these fungi were non-rock phosphate solubilizers, but two isolates, Aspergillus niger and Penicillium citrinum, had high activity. Liquid culture experiments revealed that both fungi caused a remarkable drop in pH of culture media and solubilized considerable amounts of phosphate. The effects on wheat of inoculation with vesicular–arbuscular mycorrhizal fungi and rock-phosphate-solubilizing fungi and fertilization with rock phosphate were studied in sterilized pot soils, nonsterilized pot trials and in field plot soils. Rock phosphate fertilization and inoculation with Glomus constrictum and rock-phosphate-solubilizing fungi (A. niger and P. citrinum) significantly increased dry matter yield of wheat plants under all experimental conditions. However, the effect was more evident in non- sterilized pot soils and in the field than in sterilized pots. Rock phosphate had no significant effect on the total phosphorus content of plants grown under pot conditions but it was significantly increased in field plots; the effect of inoculation with fungi (G. constrictum, A. niger and P. citrinum) on plant phosphorus was closely related to this in dry matter production. The greatest positive effect on growth and phosphorus contents of wheat plants was recorded in the treatments that received rock phosphate and were inoculated with a mixed inoculum of the three microorganisms used, followed by dual inoculation treatments of G. constrictum plus either A. niger or P. citrinum.

The retention of phosphate (P) in soil limits its mobility through bothdiffusion and mass flow. Soil P retention is a complex reaction with aluminum(Al) and iron (Fe) playing major roles in P retention in acid soil. This studyexamined the one-dimensional diffusion of applied phosphate andwater-extractable Al in a gibbsite-rich acid soil as affected by liming. Thesoil used was a loamy sand, a Typic Hapludox (US Taxonomy), obtained from asite150 km south of Perth, Australia. Soil was limed at 5 gkg–1 to raise the soil pH from 5.5 to 7.1.KH2PO4 was applied to the surface of unlimed and limedsoil columns, which were then incubated in a constant humidity chamber for 3,7,14 and 28 d. Water-extractable P (WE-P) concentration in the surface ofunlimed soil (0 to 0.2 cm) increased from 0 in the original soilto22.3 mol g–1, 3 d after the Papplication. However, the WE-P decreased with incubation period to a value of9.7 mol g–1 on day 28. Similar WE-Pconcentration patterns were observed with the limed soil, but the WE-Pconcentration near the surface decreased to a much lower value under limed (7.6mol g–1) than unlimed (9.7 molg–1) conditions. Liming also reduced P diffusion. After28 d of incubation P diffused to a depth of 2.1 cmin the unlimed soil, but to only 1.5 cm under the limedsoil.Acid-extractable P (AE-P) also increased from 0 to 50 molg–1 in the surface soil layer for both unlimed and limedsoil as a result of P fertilizer application. There was little change in theAE-P for both treatments from day 3 of incubation to day 28. The pH and Al andFe concentrations in the soil solution were elevated near the soil surfacewhereK and P ions interacted with soil matrices. Liming significantly reduced thetotal amount of WE-P and P diffusive movement in the soil, but reduced Altoxicity since Al ions shifted to less toxic species with increases in pH. Pfertilizer application also increased soil pH under both unlimed and limedconditions. This would reduce Al toxicity, benefiting crop production.
