Study of PR Gene Expression Pattern related to in Induced Resistance to Powdery Mildew in Susceptible Wheat Genotype after Treating with Salicylic Acid

Abstract Bread wheat is one of the most important food crops which has main position in nutrition of the world's population. The production of this crop were always been faced to a variety of challenges. Powdery mildew, caused by the biotrophic pathogen Blumeria graminis f.sp tritici (Bgt), known as a destructive disease of wheat worldwide. There are several methods to control the disease, but use of inducers as of SA, able to induce a systemic resistance in plants are very important. In order to examination the ability of SA for inducing resistance in wheat against Bgt fungus, Flat was selected as susceptible cultivar and treated with SA and then exposed to Bgt fungi together with control plants. Then examine the expression rate of PR1, PR2, PR3 and PR5 genes using Real Time PCR technique in response to Bgt was carried out at 5 time courses and in 3 independent replicates. Results indicated that in both groups of treated and control plants, levels of gene expression were increased after infection for all genes. Maximum expression level of genes were observed at 24 hours after infection. This process was observed slowly in control plants but caused early and faster induction of plant defense genes in treated plants early hours after infection rather than control plant. At 48 hours after inoculation, transcript levels of induced genes started to dampen in both groups of experimental plants, indicating effective suppression of defense associated genes upon haustorium development. Overall, results indicated that SA inducer beacause of able to induce resistance in susceptible cultivar through overexpression of resistance genes and reducing number of colonies grown on Bgt fungus per unit area can one of the candidate of induced resistance in susceptible cultivar. Abstract
Bread wheat is one of the most important food crops which has main position in nutrition of
the world's population.The production of this crop were always been faced to a variety of
challenges. Powdery mildew, caused by the biotrophic pathogen Blumeria graminis f.sp tritici
(Bgt), known as a destructive disease of wheat worldwide. There are several methods to control
the disease, but use of inducers as of SA, able to induce a systemic resistance in plants are very
important. In order to examination the ability of SA for inducing resistance in wheat against Bgt
fungus,Flat was selected as susceptible cultivar and treated with SA and then exposed to Bgt
fungi together with control plants. Then examine the expression rate of PR1,PR2,PR3 and PR5
genes using Real Time PCR technique in response to Bgt was carried out at 5 time courses and
in 3 independent replicates. Results indicated that in both groups of treated and control plants,
levels of gene expression were increased after infection for all genes. Maximum expression
level of genes were observed at 24 hours after infection. This process was observed slowly in
control plants but caused early and faster induction of plant defense genes in treated plants early
hours after infection rather than control plant. At 48 hours after inoculation, transcript levels of
induced genes started to dampen in both groups of experimental plants, indicating effective
suppression of defense associated genes upon haustorium development.Overall, results
indicated that SA inducer beacause of able to induce resistance in susceptible cultivar through
overexpression of resistance genes and reducing number of colonies grown on Bgt fungus per
unit area can one of the candidate of induced resistance in susceptible cultivar.

Barley is one of the most important cereals in animal nutrition and it has a role as a substitute source of wheat and rice in diet nutrition. This product is always exposed to damaging factors such as pests and diseases which lead to reduce yield and quality. The Powdery mildew agent, as a biotrophic fungus, seriously causes damage in some barley plantation areas. Similar to other plants, barley employs diverse mechanisms against the disease based on proteins and other antimicrobial agents such as pathogenesis- related proteins which have essential roles in disease resistance. In this study, morphological and molecular analyses were conducted on some barley genotypes after challenge inoculation with Blumeria graminis f.sp hordei (Bgh), the causal agent of powdery mildew. For this purpose, 11 barley genotypes were selected for initial screening. Result from data analysis showed that Mb-86-5 was the most susceptible cultivar and in Avt/Attiki, Ceres, BIR, L.1242, Yousef, Nosrat, Mahoor, Comp-1-71E and Rihane-03 genotypes, the rate of Bgh colony number significantly decreased when compared to susceptible genotype, The susceptibility rate between Beecher and Mb-86-5 was not significant. Determination of the genes expression involving in disease resistance, carried out on one- week old seedling. For all samples total RNAs were extracted. Molecular investigation showed that PR1, PR5, NH1, POX genes enhanced significantly in resistant genotype Ceres when compared to Mb-86-5 susceptible genotype. Results of this study indicate that the mentioned genes are involved in powdery mildew disease resistance in Ceres barley genotype. Infection of plants with pathogens like viruses or fungi often leads to a response of the host ranging from very mild to very severe. A very mild response usually does not result in considerable changes in the patterns of gene expression of the plant. However, a hypersensitive reaction, e.g., in the case of infection of Samsun NN tobacco with tobacco mosaic virus, is accompanied by a massive increase in the amounts of a large number of different proteins. Among these induced proteins are members of a group of so-called pathogenesis-related (PR) proteins. Frequently, the hypersensitively reacting host becomes resistant to subsequent pathogenic attack. The correlation of this phenomenon of acquired resistance and the induced protein synthesis suggests a direct involvement of these proteins in the observed replication deficiency of the pathogen. This has initiated studies by several research groups toward a better understanding of the role of these PR-proteins in the hypersensitively reacting plant. This review deals with the progress that has been made with the analysis of the host response and the function of the PR proteins in the acquired resistance.

The effect of powdery mildew (Blumeria graminis f. sp. tritici) on the grain yield and protein content of one susceptible, Springfield, and three moderately resistant cultivars, Fielder, AC Reed and AC Nanda, of soft white spring wheat (Triticum aestivum) was examined at two field locations near Lethbridge and Vauxhall, Alberta, in 1999 and 2000. At the start of heading, powdery mildew development was suppressed in half of the plots of each cultivar by a single spray application of the fungicide Tilt (propiconazole). Severe powdery mildew infection of the susceptible cultivar Springfield resulted in yield reductions ranging from 11.4 to 19.9%. The grain yield of the moderately resistant cultivar Fielder was significantly reduced at both sites in 1999 by 7.6-10.5% while AC Reed suffered a significant yield loss (7.6-9.1%) at Lethbridge in both years. The moderately resistant cultivar AC Nanda consistently had the lowest powdery mildew ratings and its yield was unaffected by the disease. A single fungicide application prevented disease buildup on the moderately resistant cultivars, but not on Springfield. The grain protein content of the moderately resistant cultivars was unaffected by powdery mildew, but it decreased in Springfield by 0.6-0.7%.

A field study was undertaken to ascertain the effects of elicitors viz. benzothiadiazole (BTH) and salicylic acid (SA) on defense related enzymes viz. peroxidase, phenylalanine ammonia lyase, superoxide dismutase, chitinase and β-1,3-glucanase, and phenols in rice (Pusa Basmati I) plants. First foliar spray of BTH (50 mg/kg) and SA (50 mg/kg) was done at the maximum tillering stage and inoculation with Rhizoctonia solani was carried 24 h after elicitor treatment. Elicitors were further sprayed at every growth stage. Time course analysis showed peak accumulation of defense related enzymes and phenols in the rice leaves treated with BTH and SA, and accumulation was the highest at the flowering stage. Higher enzymatic activity was observed in elicitor treated plants inoculated with R. solani. Compared to the untreated control plants, application of elicitors before R. solani inoculation significantly elicited the defense related enzymes and phenols. Moreover, application of elicitors had a positive effect on yield and disease reduction. It is suggested that pretreatment of rice leaves with BTH and SA could be used to enhance the level of protection against sheath blight and to improve rice yield in the fields.

This review covers the physiology and molecular biology of the plant beta-glucanases possessing either endo-1,3-beta-D-giucanase (EC 3.2.1.39) or endo-1,3;1,4-beta-D-glucanase (EC 3.2.1.73) activity. These beta-glucanases are structurally related enzymes that are believed to be involved in many important aspects of plant physiology and development, such as germination, growth, defense against pathogens, flowering, cellular and tissue development and differentiation, and probably other roles. They also are regulated by numerous plant hormones, biotic and abiotic elicitors and stresses, and they exhibit complex tissue- and developmental-specific gene expression.

Recent applications of techniques in plant molecular biology and biotechnology to the study of host–pathogen interactions have resulted in the identification and cloning of numerous genes involved in the defense responses of plants following pathogen infection. These include: genes that express proteins, peptides, or antimicrobial compounds that are directly toxic to pathogens or that reduce their growth in situ; gene products that directly inhibit pathogen virulence products or enhance plant structural defense genes, that directly or indirectly activate general plant defense responses; and resistance genes involved in the hypersensitive response and in the interactions with avirulence factors. The introduction and expression of these genes, as well as of antimicrobial genes from nonplant sources, in a range of transgenic plant species have shown that the development of fungal pathogens can be significantly reduced. The extent of disease reduction varies with the strategy employed as well as with the characteristics of the fungal pathogen, and disease control has never been complete. Manipulation of salicylic acid, ethylene, and cytokinin levels in transgenic plants have provided some interesting results with regard to enhanced disease tolerance or susceptibility. The complex interactions among the expressed gene product, plant species, and fungal pathogen indicate that the response of transgenic plants cannot be readily predicted. Combinations of defense gene products have shown considerably more promise in reducing disease than single-transgene introductions. The use of tissue-specific or pathogen-inducible promoters, and the engineered expression of resistance genes, synthetic antimicrobial peptides, and elicitor molecules that induce defense responses have the potential to provide commercially useful broad-spectrum disease resistance in the not-too-distant future. The issues and challenges that will need to be addressed prior to the widespread utilization of these transgenic plants are highlighted.

The two most commonly used methods to analyze data from real-time, quantitative PCR experiments are absolute quantification and relative quantification. Absolute quantification determines the input copy number, usually by relating the PCR signal to a standard curve. Relative quantification relates the PCR signal of the target transcript in a treatment group to that of another sample such as an untreated control. The 2(-DeltaDeltaCr) method is a convenient way to analyze the relative changes in gene expression from real-time quantitative PCR experiments. The purpose of this report is to present the derivation, assumptions, and applications of the 2(-DeltaDeltaCr) method. In addition, we present the derivation and applications of two variations of the 2(-DeltaDeltaCr) method that may be useful in the analysis of real-time, quantitative PCR data.
