Karyomorphology of Turkish species in Centaurea sections Centaurea and Phalolepis (Asteraceae) and implications for taxonomy

Abstract and Figures

In this paper, 31 mostly endemic and locally distributed Turkish Centaurea taxa belonging to Centaurea and Phalolepis sections were examined in terms of their karyomorphology. The basic chromosome number for all of the studied species was concurringly determined as 9(x = 9) for both sections, excluding C. hierapolitana. We also determined tetraploid and hexaploid species in spite of many species having diploid chromosome numbers. Twelve chromosome counts are reported for the first time and most of the karyotyping analyses are described for the first time via the KAMERAM program. The karyotypes had a predominance of metacentric (m) chromosomes. However, in the karyotyping of six taxa, submetacentric (sm) chromosomes were dominant. Five quantitative asymmetric indices were used to evaluate karyological features of the species. A meaningful dendrogram was carried out to assess the karyotype-symmetry conditions and describe the karyotyping relationships between different taxa.

In this article, a new cornflower, Centaurea turhanii Uysal & Hamzaoğlu (Asteraceae) from Türkiye was described and reported. Morphological, karyological, and molecular analyses were used to reveal the taxonomic status of the new species with the species thought to be related to taxa. A new Centaurea species, C. mengenensis Uysal & Şirin, is described and illustrated from Mengen district, Anatolia, based on morphological and molecular data. To identify its taxonomical position within Centaurea, the internal transcribed spacer (ITS) = gene region of the new species was amplified and sequenced. The new species belongs in Centaurea subg. Centaurea as shown by both morphological and molecular data. Its diagnostic characters are presented, including an updated key to related species distributed in Turkey. Additionally, the geographic distribution of the new species, as well as of its relatives, is mapped. The chromosome number of C. mengenensis is 2n = 2x = 18 and its achene surface ornamentation is irregularly sulcate.

A new species of Centaurea sect. Phalolepis (Asteraceae) is described and illustrated from Turkey. This species was collected from Vezirköprü district (Samsun Province) in north Anatolia. Centaurea isilae sp. nov. is morphologically similar to C. lycia and C. cadmea ssp. pontica from which it differs mainly in basal leaves, upper leaves and achene features. Comprehensive descriptions and comparison of the new species and closely related species, ecology, conservation status, pollen morphology, a distribution map and detailed illustrations of the new species are given.

An evaluation was conducted of A. yamadagensis, which is a Critically Endangered (CR) species that is endemic to Turkey, comprising an analysis of its taxonomical, micromorphological, macromorphological, cytological and palynological features, in addition to its geographical distribution. Scanning Electron Microscopy was used to obtain seed and pollen surface microphotographs. The shape and sculpturing of the seed periclinal walls are convex and granulose. The shape of the pollen grains appears monocolpate, with polar and equatorial axes of 23.65 ± 0.29 and 14.92 ± 0.33 µm, respectively. A. yamadagensis has a pollen ornamentation that is foveolate-microreticulate. Cytological analyses revealed that A. yamadagensis is diploid and has a chromosome number of 2n = 2x = 16.

Five endemic Muscari taxa in Turkey are examined in terms of chromosome number and morphology. Muscari sivrihisardaghlarense (2n=18), M. serpentinicum (2n=18), M. atillae (2n=18) and M. ufukii (2n=18) are diploid. M. tuzgoluense is polyploid with 2n=36 chromosome number. Chromosome counts have confirmed the previous reports that Muscari genus is x=9 of basic chromosome number. The chromosome number and morphology of these species are reported for the first time. The karyotypes have the predominance of metacentric and submetacentric chromosomes. Six quantitative asymmetric indices were used to evaluate the karyological characteristics of the species, and these results increase our karyological knowledge about the species.

Chromosome numbers of five Alyssum species (A. aizoides Boiss., A. aureum (Fenzl) Boiss., A. baumgartnerianum Bornm. ex Baumg. A. constellatum Boiss. and A. xanthocarpum Boiss.) from Turkey have been reported for the first time. A. aizoides and A. baumgartnerianum are tetraploid (2n=4x=32) while A. au-reum, A. constellatum and A. xanthocarpum are diploid (2n=2x=16).

Centaurea uysalii Şirin & Çeçen, a new Asteraceae species from the province of Karaman, in Turkey, is described and illustrated herein. It is morphologically similar to C. bourgaei Boiss. (Subgen. Cyanus), but it mainly differs from its relative in terms of the involucre and achene lengths. The achene micromorphological features and karyomorphology of C. uysalii and C. bourgaei were examined in this study. The IUCN conservation status is the Critically Endangered [CR B2 a b (i, iii)] category.

Centaurea osmaniyensis İlçim (Asteraceae) sp. nova is described and illustrated from Bahçe District, Osmaniye Province, Turkey. The species belongs to sect. Cynaroides and morphologically closely resembles C. regia var. cynarocephala, but differs from it by the habit, petiolate leaves, stem and leaf indumentum, capitulum shape, phyllary length, appendage size, terminal spine length and pappus color. The seed and pollen micromorphology are presented, as well as the threat level is evaluated.

Özet: Bu çalışmada, Centaurea pseudoscabiosa türüne ait alt taksonların altı farklı popülasyonunun karyotipleri aseto-orsein yöntemi ile incelenmiştir. Tüm popülasyonlar aynı kromozom sayısına (2n=22) ve temel kromozom sayısına (x=11) sahiptir. Alt taksonların karyotipleri C. pseudoscabiosa subsp. pseudoscabiosa ve C. pseudoscabiosa subsp. araratica 22m, C. pseudoscabiosa var. sipikorensis, 20m+2sm ve C. pseudoscabiosa subsp. glehnii 16m+ 6sm olarak tespit edilmiştir. Alt taksonların karyolojik özelliklerini değerlendirmek için beş kantitatif asimetrik indeks kullanılmıştır. Asimetri indekslerine göre, C. pseudoscabiosa subsp. glehnii en asimetrik kromozomlara sahiptir (CVCI:10, AI:1,8). Taksonomik açıdan karyolojik veriler C. pseudoscabiosa’nın önceki tür altı sınıflandırmasını doğrulamaktadır. Ayrıca karyolojik sonuçlar, Türkiye Florası’nda sinonim olarak kabul edilen C. pseudoscabiosa var. sipikorensis taksonunun alt tür olarak yeni bir taksonomik düzenlemeyle canlandırılması gerekliliğini desteklemektedir. Abstract: In this study, karyotypes of six different populations of sub-taxa belonging to Centaurea pseudoscabiosa Boiss. & Buhse were investigated by aceto-orcein method. All populations have the same chromosome number (2n=22) and basic chromosome number (x=11). Karyotypes of sub-taxa C. pseudoscabiosa subsp. pseudoscabiosa and subsp. araratica, C. pseudoscabiosa var. sipikorensis, and C. pseudoscabiosa subsp. glehnii has been identified as 22m, 20m+2sm and 16m + 6sm, respectively. Five quantitative asymmetric indices were used to evaluate the karyological characteristics of sub-taxa. According to the asymmetry indices, C. pseudoscabiosa subsp. glehnii has the most asymmetric chromosomes (CVCI:10, AI:1.8). From a taxonomic point of view, karyological data confirm the previous subspecies classification of C. pseudoscabiosa. In addition, the karyological results support the necessity of reviving the taxon C. pseudoscabiosa var. sipikorensis, which is accepted as a synonym in the Flora of Turkey, with a new taxonomic arrangement as a subspecies.

In this study, the karyomorphology of five endemic Arabis taxa (A. alanyensis, A. androsacea, A. aubrietioides, A. davisii, and A. deflexa) was examined. The chromosome number of these species was determined for the first time. As a result of the karyomorphological studies, the chromosome number was determined to be x =8 for all species. Arabis androsacea and A. aubrietioides are tetraploid whereas other species are diploid. Considering the asymmetry indices, we can conclude that all taxa have symmetrical karyotypes. The most common karyotype formulas are 2n = 4x = 32m and 2n = 2x = 14m + 2sm, respectively. Arabis davisii (on the second chromosome pair) and A. deflexa (on the third chromosome pair) are distinguished from others by showing a secondary constriction.

Centaurea baseri, a new species from limestone rocks in Emirdaǧ (Afyon), Central Anatolia, Turkey, is described and illustrated. The new species is compared with the most similar species C. aphrodisea and C. dursunbeyensis (C. sect. Phalolepis), from which it differs in its involucre, basal leaves, achenes, appendages and habit. Diagnostic morphological characters are discussed. Notes are presented on its ecology and conservation status.

Centaurea L. (Asteraceae) is one of the largest genera in Turkey. The chromosome number and morphology of 8 taxa belonging to the genus Centaurea were investigated using an image analysis system. Among these taxa, the chromosome numbers were as follows: 2n = 18 in C. cariensis Boiss. subsp. niveotomentosa (Hub.-Mor.) Wagenitz, C. lycaonica Boiss. & Heldr., C. virgata Lam.. and C. Polyclada DC.; 2n = 24 in C. cyanus L.; 2n = 36 in C. virgata and C. cariensis Boiss. subsp. cariensis: 2n = 40 in C. urvillei DC. subsp. urvillei; 2n = 54 in C. tuzgoluensis Aytac & H.Duman. In 5 taxa mixoploidy was observed (C. lycaonica, C urvillei subsp. urvillei, C cariensis subsp. cariensis, C. virgata. and C. tuzgoluensis). Based on karyotypic data for this genus, 3 basic chromosome numbers in Centaurea taxa were observed (x = 9. 10. and 12). Furthermore, ideograms are given for all the Centaurea taxa studied.

Reports of 89 new counts are recorded for the tribe Helenieae, including reports for 28 genera and 56 species. One genus, Hypericophyllum (n = 9), and six other taxa were previously unreported.

In several systems of the genus Centaurea the sections Acrolophus and Phalolepis have been placed in different subgenera, because of the striking differences in the shape of the appendages of the phyllaries. Despite this they agree closely in chromosome-number, pollen-morphology, carpology and in general habit. It is shown that several species pairs exist — one species traditionally in section Acrolophus, the other in section Phalolepis —which agree in most characters and live in adjacent areas. Their close affinity can not be denied. The conventional recognition of the 2 sections is unnatural.

Centaurea wiedemanniana is an endemic plant species native to Turkey. The plant is known to have a narrow distribution in two abutting counties, Bilecik and Eskisehir. Genetic variability within and among 6 known natural populations of this plant was assessed by using RAPD markers. High levels of genetic diversity were detected both at population (PPB = 62.83%, I = 0.301, H-S = 0.196) and species level (PPB = 94.65%, I = 0.429, H-T = 0.278). Analyses of molecular variance showed that among and within-population genetic variation accounted for 71.70% and 28.30% of the total genetic variation, respectively. A high degree of genetic differentiation was also found among populations (G(ST) = 0.223). The genetic structure of C. wiedemanniana has probably been shaped by its breeding characteristics, habitat preference and short distance seed and pollen dispersal. The observed genetic variations suggest that as many populations as possible should be considered in any planned in situ or ex situ conservation programs for this vulnerable endemic species.
