Clonal propagation of Humboldtia vahliana Wt. - An endemic tree of Southern Western Ghats

Abstract

Humboldtia vahliana Wt., (Fabaceae - Caesalpinioideae) an economically important endemic tree of Southern Western Ghats is found susceptible to seed infestation and subsequent poor seedling bank in their natural habitats. This foreseen untimely endangerment of the species. The present study is therefore aimed to establish the clonal genetic stock of the species by inducing rooting in different aged stems by using auxins IAA, IBA and NAA with concentrations of 250, 500,1000 and 2000ppm. The stem cuttings from 1-2 year old juvenile plants resulted maximum rooting of 90% along with 80% survival of the ramets by the use of IBA at 1000 ppm. Thus, the standardized clonal propagation method in H. vahliana through juvenile stem cuttings reported first time could be taken as an ideal tool for the multiplication and subsequent resource base utilization of the species since the populations are prone to endangerment in its habitats.

Coscinium fenestratum (Gaertn.) Colebr., a critically endangered medicinal plant of Western Ghats, is a potential substitute for Berberis chitria because of its berberine content. The destructive harvest of the plant for both stem and root has resulted the paucity of reproductive individuals in their distributional range and subsequent arrest of natural regeneration. The present study is therefore conducted to raise clonal genetic stock of the species by inducing rooting in different aged stem cuttings and through air layering using auxins such as IAA, IBA and NAA with concentrations ranging from 250-2000 ppm. The juvenile plants of 2-3 year old has resulted a maximum stem rooting of 60% and air layering success to 90% by the use of NAA at 1000 ppm. The clonal propagation methods standardized in C. fenestratum is a first report of its kind and could be an ideal propagation technique for the mass multiplication that will lead to conservation and sustainable utilization of this rare species.

Clonal propagation through stem cuttings and air layering in Dysoxylum malabaricum Bedd. ex Hiern. is presented here. The highest rooting of 80% was achieved with the use of IBA -1000 ppm from tender stem cuttings of 10-15 years old plant and air layering success was also found to be 90% by the use of IBA 1000 ppm from same age of plants used for stem rooting.

Detailed studied were conducted on the efficacy of clonal and seed germination behaviour of an endemic leguminous tree (C.b.) in Western Ghats. The study revealed maximum stem rooting (75%) and air layering success (60%) by the use of IBA 3000 and IBA 1000ppm respectively. The moderately recalcitrant type of seeds with 51% moisture content had shown 100% germination and 40% when attained critical moisture content at 38 to 40%. Overnight water soaking influenced early germination (5-6 days) than control sets. The present study is therefore first time optimize clonal and seed propagation methods of the species further to raise clonal genetic stock and germplasm storage for effective ex situ conservation from its local extinction.

The juvenile plants of Drypetes malabarica have resulted 10070 rooting success for both stem cuttings and air layering by the application ofNAA 3000 and IAA l000lpm respectively. The rooted plants were also survived with cetrt p€rcent success rate. Since the tree is being prore to high degee offrtrit predation, seed infestation and overall poor natural regeneration iz sir|l restricts, the soil seed and seedling baok and merit extend cloml propagatio[ of the species. The propagation methods thus developed fust time could be adopted for the large scale multiplication, establishing clonal gene banks er situ which in tum ensures the availability ofmaterials for conservation, evaluation and management pnctices lvithout affecting natural populations ofthe species in the long run.

Tabernaemontana gamblei (Apocynaceae), an important endemic shrub of the southern Western Ghats is found very poor seedling bank in their natural habitats. This will lead to the untimely endangerment of the species in near future. The present study is, therefore, aimed to develop and standardise the macropropagation techniques through stem cuttings and air layering methods with the aid of rooting hormones. Ten year old mother plants have resulted a maximum stem rooting of 70% in the nursery conditions and air layering success of 60% in their natural habitats, by the application of NAA 1000 ppm in both the cases. The plants developed from stem cutting and layering were also survived with 80% success. Thus the standardized clonal propagation method in T. gamblei through stem cuttings and air layering reported for the first time can be taken as an ideal tool for the multiplication and subsequent resource base utilization of the species since the populations are prone to endangerment in its habitats.

The highest rooting of 90% was achieved with the use of IBA 1000 ppm from tender stem cuttings of 15-20 years old plants while air layering was found to be 100% by the use of IBA 1000 ppm. Thus the standardized clonal propagation method is recommended as potential methods for multiplication, conservation and management of these endangered and red listed tree resources from untimely endangerment for the posterity.

The paper deals with the effect of six rooting formulations including control on rooting behaviour of cuttings from juvenile chir stock. Cent per cent rooting was recorded when cuttings from 1.5 years old ortete were treated with 0.5% IAA-talc and stuck in mist unit during June.
