Abstract

The present study was conducted to evaluate the anti-inflammatory, analgesic and anti-arthritic activities of Mahanarayana tailam (MT) in laboratory animals. Carrageenan induced rat paw oedema, Freund's adjuvant induced arthritis in rats and Eddy's hot plate induced pain in mice were used as experimental models. MT was applied to the planter surface of the hind paw by gently rubbing to treatment groups. The external application of MT, showed significant inhibition of inflammation (P < 0.01) as compared to control and less than Piroxicam gel 0.5%. Analgesic effect as compared to control was found to be significant (P<0.01) and almost equal to the standard drug. Anti-arthritis activity of the oil was observed almost equal to the standard drug used in the study. Histopathology of ankle joint revealed that Piroxicam as well as MT inhibits the inflammation in the treated animals. However, mild mononuclear infiltration of surrounding tissue was observed in the MT treated animals. These observations suggest that MT may be used to treat inflammation, pain and arthritis. It is known that all conventional analgesics and anti-inflammatory agents produce various adverse effects. In an attempt to avoid adverse gastrointestinal effects, research is now focusing on Ayurvedic drugs which are devoid of such adverse effects.

Fritillaria roylei (Kshirakakoli) is a primal plant used in ancient times. But nowadays, due to biotic and abiotic stress the plant has entered in the list of threatened medicinal plant. In ancient texts effective uses of formulations containing kshirakakoli are well mentioned but the information is not written in simple language due to which the therapeutic value of the plant is not well understood by scientific fraternity. So, there is a major need to perform ethno medicinal survey for the formulations containing kshirakakoli and compare their therapeutic uses as mentioned in text with the modern system of medicine. In this study, a field survey was performed in 4 states i.e., Uttar Pradesh, Uttrakhand, Punjab and Himachal Pradesh where the conversation regarding usage of this plant or formulation was done with 24 local medical practitioners, 18 shopkeepers and 4 traditional healers. The information thus obtained were recorded and then compared. Results showed that the usage of kshirakakoli containing formulations was highest in Himachal Pradesh and Uttrakhand. Only few clinical studies have been done on these formulations. The effectiveness of the formulations against remedies alluded by the trado-medical practitioners claimed was found accurate as per ayurvedic textual literature. Hence, the ethno medicinal survey provides a precise guidance to scientists for future research on these kshirakakoli containing formulations that are useful in plethora of disorders.

The present study describes an approach to synthesize the smaller chain tripeptides and tetrapeptides and to test their antinociceptive potency in mice. Based on rational drug design using hydrophobic ratio and total net charge as descriptors, five leads were selected, viz., Met-Arg-Tyr (MRY), Met-Val-Tyr (MVY), Met-Ile-Cys-Tyr (MICY), Met-Trp-Lys-Tyr (MWKY) and Phe-Trp-Lys-Tyr (FWKY) from the subjected 65 templates and synthesized by dicyclohexyl carbodiimide coupling using polystyrene as solid support. All the synthesized compounds were purified by column chromatography and further confirmed by melting point, infrared, proton nuclear magnetic resonance and mass spectral datas. Acute toxicity studies were performed for dose selection in all the compounds using OECD guidelines 423 (Annexure 2b). Antinociceptive potency of peptides was tested in Swiss albino mice using acetic acid writhing and hot plate method. The LD50 cut-off mg/kg body weight for tripeptides (MRY, MVY) and tetrapeptides (Met-Ile-Cys-Tyr, Met-Trp-Lys-Tyr, Phe-Trp-Lys-Tyr) were found to be 500–2000 mg/kg and 200–300 mg/kg respectively. The tripeptide MVY have shown maximum antinociceptive action with an average number of writhing as 13 at 150 mg/kg body weight by writhing method, whereas the tripeptide Met-Arg-Tyr have shown maximum potency with the average reaction time of 5.75 min after 15 min at a dose of 150 mg/kg by hot plate method, which clearly indicated that the tripeptides are comparatively potent antinociceptive agents than the tetrapeptides for central neuropathic pain.

Due to role of nutraceuticals in health care, herbal products have become major concern for food industry and health-care regulatory bodies. The World Health Organization estimates that Ayurveda is gaining popularity due to its easy availability, low cost, congeniality, and higher safety. Ashtawarga group of plants used in a number of aphrodisiacs Ayurvedic formulations/functional foods have been included in the Red List of Threatened Species. To overcome this problem, permission to use official substitutes is being exploited by manufacturers resulting in serious problem of adulteration with spoiled, inferior, and spurious drugs. Plant dealers have discovered the “scientific” methods to adulterate a high-quality material, which is difficult to analyze without high-end equipment and chemical markers. Confirmation of text-based use of these plants may further throw light on medicinal and therapeutic potentials of these eight plants and could be a great step toward high-quality nutraceuticals in future.

Fenugreek also called as Trigonella foenum graecum, belong to Fabaceae family. It is one of the most common Indian spices that are particularly recognized for its culinary, medicinal, and aromatic properties. Fenugreek is more pronounced for its health-promising properties such as antiinflammatory, antiulcer, antidiabetic, and anticancer activities. Fenugreek is enriched with various chemical constituents that aids in enriching its nutraceutical properties. Fenugreek is enhancing human health by providing different bioactive compounds, antioxidant compounds, and hepatoprotective capacity. Fenugreek gel formation property aids in preventing antiulcer as studied with respect to some antiulcer drugs available like omeprazole. Fenugreek is one major kitchen ingredient that controls various metabolic diseases by its different nutraceutical properties. Based on the past evidences, the present review chapter aims to elaborate the nutraceutical properties of fenugreek.

INTRODUCTION India from times immemorial has been blessed by nature with the wealth of fascinating and alluring medicinal plants. In the oldest repositories of human knowledge, plants and plant products have been positively recognized as supportive agents in the treatment of various diseases. [1] Ayurveda was not in theoretical form but was passed on from generation to generation by teacher in the practical form till the Ayurveda was compiled and found its rightful place at the end of the fourth Veda, the Atharvaveda. Due to a different Study Background: Roscoea purpurea (Kakoli) is a wonder plant used by Saints/Rishies since ages, however, entered the list of plants facing extinction due to enumerable reasons. Ancient texts claim very potent uses of kakoli-containing formulations, but the information is available in regional languages and hence the real usages of the plant are not well understood by scientific fraternity. Hence, it becomes important to know the real status of the mentioned therapeutic potentials as well as practiced potentials by tradomedical practitioners. Hence, the major objective of the work was to conduct the ethnomedicinal survey for intended formulations and compares it with therapeutic usage mentioned in text/modern system of medicine. Materials and Methods: A field survey was conducted in Himachal Pradesh, Punjab, Uttrakhand, and Uttar Pradesh followed by discussion with 18 shopkeepers, 24 local medical practitioners, and 4 traditional healers. The information regarding usage of these formulations were recorded and compared. Results: Results showed that usage of formulations containing kakoli was highest in Himachal Pardesh and Uttrakhand. Only few clinical studies have been done by scientists on these formulations. Conclusion: The efficacy of the remedies alluded by the respondents claimed to be exact as per the Ayurvedic textual literature. This survey provides a template for scientists for further screening and research on these formulations useful in plethora of disorders.
