I would like to study the aged T2DM mice and I have 12-month-old mice now. Is it possible to induce them T2DM by HFD+STZ? Most studies induce T2DM from 8 weeks old, but it's too long to wait until old. Some people inject old mice with STZ to induce T1DM, but I didn't find studies that use both STZ and HFD in 12month old mice.

Hi everyone,
I am trying to differentiate PC12 cells in 96 well plate ( 10000/ well) with 50ng/ml NGF.
In the third day, some cells have outgrowth. However, cells kept on dividing and forms clump. Is this normal? will they keep divide during differentiate? Maybe the seeding density was high?

I want to treat PC12 cells with high glucose and palmitate after it differentiated. First, I will differentiate PC12 cells on 24 plates. After differentiation, can I frozen PC12 cells or change to other plates for glucose treatment?

I'd like to exposure PC12 cell with both hyperglycemia and palmitate, then test apoptosis and oxidative stress, to study diabetic neuropathy.
PC12 need NGF to induce differentiation, when do I need to add NGF or induce differentiation, before I exposure the cells to hyperglycemia and palmitate or add high glucose and NGF in the meantime?

Hello,
My project is trying to examine the effect of a bioactive component on diabetic-induced brain damage. we have high-fat high sugar induced diabetes mice model, then we do some analysis of mice brain. At the mean time, I want to mimic a same condition in cell model, to do other analysis. The cell lines could be SHSY5Y or PC12 cell line. Does anyone has any idea?

I have 5 treatment groups male mice sample and 5 treatment groups female mice, and I wanna to do western blotting to see if there any sex-dependent difference among markers (I need to compare male and female).
When I load the samples, do I have to load male and female in the same gel? I only have 20 wells in one gel, if I choose 2 samples for each groups, I do not have space to load markers. Can I just choose one samples from each group? or Can I load female and male in separate gel?
