What is a good method for Visualizing Endophytes?

How do I prepare a segment of vascular plant stem for free hand cross sectioning? I want to make slides of thin cross-sections to look for varying concentrations of fungal hyphae at different points along a continuous plant stem.

 You may try free hand sectioning with razorblade, or embed in a block of 3% agar for additional support if you cut thin stems. It is better to use fresh tissue, but it works with fixed tissue also. Sections of up to 50um can be used for subsequent staining.
