What are the values needed to calculate the total phenolic content?

Since the TPC formula is cv/m, i want to know what sample volume v refers to. Is it the volume of the diluted sample or the sample we mixed when conducting the TPC assay?

Most recent answer

the sample volume (v) refers to the volume of the diluted sample that was used in the TPC assay. It is important to accurately measure and report the sample volume used, as it is a factor in the calculation of the total phenolic content.

for the evaluation of phenolics content : total phenolic compounds (TPC). The results were given as milligram of acid Gallic per gram of dry powder (mgGAE/G.dw). 20 μL of the sample was added to 100 μL of Folin-Ciocalteu reagent in addition to 80 μL of Na2CO3 , the mixture was incubated for 10 min and the absorbance was performed at 765 nm (Atanassova.,2005). for more details, see Sustainable Chemistry and Pharmacy 31 (2023) 100933 4 S.O. Hello, Total phenolic compounds (TPC) were analyzed using a UV–Visible spectrophotometer at 765 nm, and the results were given as milligram of acid Gallic per gram of dry powder (mgGAE/G.dw). 20 μL of the sample was added to 100 μL of Folin-Ciocalteu reagent in addition to 80 μL of Na2CO3 , the mixture was incubated for 10 min and the absorbance was performed at 765 nm (Atanassova.,2005). see the recent article Sustainable Chemistry and Pharmacy 31 (2023) 100933 4 S.O.
