I need your comments on the optimal conditions for measuring underivatized N-glycans in ESI-MS in negative mode?

N-glycans purification procedure and parameters in mass spectrometer.

Hi Gleysin,
I have made good experiences with Hypercarb Columns (Thermo) for the analyzis of underivatized Glycans. You can find an application note for glycopetides here (page 10ff):
In my xperience sometimes positive mode will lead to better results even in glycan analysis. If you have access to an Thermo QExactive mass spectrometer you can measure pos and neg mode simultanousely which will give you the best results.
