How to analyse the TNBS assay?

I am conducting TNBS assay to determine the number of the primary amine group of a protein. Based on the protocol, I will need to do this assay with my sample and compare with an amino acid. I would like to ask about how to determine the number of amine group per molecule in my protein based on a comparison with an amino acid?

It is assumed that 1 mol of amino acid leads to the same signal as 1 mol of amino groups (mainly epsilon-amino groups of lysine) in your protein. Depending on the reaction conditions, you may get only the number of accessible amino groups.
