How see glycolysis inhibition in cancer cells ?

Topoisomerase inhibitors comprise important cancer drugs e.g. etoposide that targets human type IIA topoisomerases (Top2α and Top2β) causing errors in DNA synthesis leading to the destruction of cancer cells. As such, by trapping the cleavage complexes of the type IIA topoisomerases, etoposide effectively blocks DNA re-ligation, thereby causing DNA strand breaks that, in turn, disrupt the entire process of cell replication and preferentially promote the apoptotic destruction of cancer cells. How does it avoid destruction of the normal cells as that would impact the patient health negatively?

I have been growing mouse organoids and they grow well at first. I am using the general media and conditions suggested by Clevers´lab and other labs. Before I provide more in detail information I would like first to share the picture of how the organoids look like after the first week, when they lose the cystic morphology and acquire this blob-like morphology, every day more cells acquire these big blobs and stop growing normally (medium does not turn yellow-ish). Have anyone experience this issue or have observed this phenotype before?If so, any suggestions about what may be causing it?

I checked the literature in detail to compare different cell viability, cytotoxicity studies to check the biocompatibility of biomaterials. In some articles, they use the 'leachable-conditioned medium' method to check the biocompatibility. For this purpose first, they incubate biomaterials in cell culture medium without FBS after that they use this 'conditioned medium' with FBS in cell seeding. I am wondering in this first step why they don't add FBS directly to cell culture media and is it important for this procedure. I also saw different methods including FBS.

In BRENDA I have found the Kcat and KM values for ATP and glucose for hexokinase. In this bisubstrate reaction, how can I compute the reaction rate if I know enzyme, ATP and glucose concentrations? The problem I have is that I need to get Vmax from somewhere but for bisubstrate reactions I cannot compute Vmax from Kcat (because there are two).

Is computing Vmax as the enzyme concentration multiplied by the lowest Kcat an option?

Am I making a mistake by using Michaelis-Menten kinetics?
