How to measure neurotransmitters with HPLC?
Hi everyone,
Please can somebody help me? I would like a protocol and a method how to measure the neurotransmitters using HPLC (adrenaline, noradrenaline, serotonin, dopamine, glutamate)

f you consider selectivity and sensitivity, HPLC coupled to electrochemical detection would be method of choice for the analysis of neurotransmitters in rat/mice brain. Concentration levels up to 50 pmol/L and lower are achievable with only 5 uL sample volume injected. So it can be used in both micro dialysis experiments as well as the analysis of brain tissue (homogenates). See for example:
For neurotransmitter amino acids like GABA and Glutamate you can also use HPLC-ECD, using an automated derivatisation procedure (done by the autosampler) with o-phthalaldehyde (OPA). With such set-up you can achieve concentration detection limits below 10 nmol/L for GABA using 1.5 - 5 uL sample injections. See for example:

Please find attached, an HPLC method which may be used to determine some neurotransmitters, by the use of electrochemical detection.

Aslamoalaikum! Do you want to measure the neurotransmitters in whole brain or regions of brain?

1. For glutamate, it is better to use Kits if you can afford.
2. For Adrenaline, I am not sure of method as we do not estimate.
3. For Noradrenaline, serotonin, dopamine, kindly download attached pdf file.
You are requested to rephrase the sentences used in the method for estimation of biogenic amines as I have taken it from my thesis which is not yet submitted.
