How can I improve the encapsulation efficiency of my fructose-based exopolysaccharide which is used to encapsulate a hydrophobic drug?

In my case, when I increase the initial amount of drug, first the loading percentage goes beyond 100 %, and then getting less to 30 %. I want to know, whether it can happen or there is something wrong with my analysis.

Hello, I am currently researching how to encapsulate bitter hydrophobic drug-using pH-sensitive Eduragit E polymer via nanoprecipitation method. I am currently stuck on deciding how to prepare a drug-loaded nanoparticle. My plan is to dissolve the drug and polymer in acetone, prepare the aqueous phase with PVA and I will mix them very slowly for the nanoprecipitation to occur. My question is how do you know that the drug particles are entrapped by the polymer? Thanks in advance.

The samples that we're using consist of a hydrophobic drug compound and a nanopolymer (PEG-siPMNT). We synthesized it by dialysis in DW as the following step: dissolve drug in DMF, then add the polymer and TEOS, mixed well. Following by loading into the dialysis bag. DW was changed in 4hr, 8hr, 12hr and 18hr. Collected samples were supposed to be a water soluble nanoformulated solution. Instead, we yieled a gel-like state (as shown).

We wanted to be clear on which factor caused the gelation. Many attemps before yield expected results, but the gelation has become more frequent from time to time.

Does the drug have to be soluble to bind the drug to the nanoparticles?

after the interaction of drug the enhancement in the SPR band was observed the intewraction was proved by FTIR and AFM the AFM resiults shows aggregation of NPs the of NPS increases from 40nm to 200 studied bty AFM i also study time depedent interaction of drug and nanoparticles the DLS spectra showing abrupt results how to interpret the results.

I am working on encapsulating drug in Pre-PGS by solvent evaporation method. In which organic phase is PGS dissolved in THF, drug in water with tween 20. I am getting little particles at end. But my FTIR anlysis is showing hydrolysis of ester bond. No ester bond peak in final construct.

What should I do to prevent hydrolysis?

Curing PGS is not an option as it will make it insoluble...........

After synthesis calcium alginate nanoparticles, How can I measured the loading capacity from EE% and the pre-weighted row materials. Because I obtained very small amount of prepared nanoparticles after centrifugation.
