Can you help me with a western blot experiment?

I need some suggestion/advice/reference for an experiment I am planning but before I ask a question, I will provide some information related to the work:
1. I am using drug loaded nanoparticles in an invitro co-culture models consisting of smooth muscle cells, endothelial cells and THP-1 monocytes.
2. The nanoparticle is monocyte targeted
3. We hypothesize that the drug (statin- antinflammatory) loaded nanoparticles will reduce inflammation (we have done ELISA and have seen a reduction in cytokine levels compared to control).
4. Now I want to do a western blot to detect NF-kB and NLRP3 inflammasome as we want to show that there are changes at the transcriptional level also.
Question:
1. Do you think point 4 above is a good experiment to do? Will it add quality information to the results I already obtained from ELISA?
2. Since my nanoparticles are only monocyte targeted, should I do the western blot using only a monolayer of monocytes?
3. Will western blot show me a reduced NLRP3 or NF-kB if there is inflammation reduction?
I would really appreciate if anyone can answer this and/or help me plan the study or provide a reference where people have done this before.
