Persimmon (Diospyros kaki Thunb) fruit is one of the most important fruit and have been cultivated from ancient times in Korea. In this study, we found 16 EST-SSR markers that contained one or more EST-SSR sites from 246 cDNA sequences. The developing of EST-SSR marker analysis from 42 persimmon cultivars was compared by genetic relationships and morphological relationships using 6 qualitative traits (fruit related 6 traits) and 19 quantitative traits (flower related 19 traits). In this study, 25 primer sets were tested to identify PCR polymorphism and 14 potential EST-SSR primer pairs were selected. The result of morphological relationship EST-SSR marker analysis showed that the coefficient 0.02 was difficult to categorize in several groups. And then, coefficient 0.77 of genetic relationship showed that the group was classified as four groups. The result of correlation distance between genetic relationship and morphological relationship were investigated was low significance (-0.03). Our results also provided an optimized method for improvement of breeding efficiency and introduce of superior character at persimmon cultivars using EST-SSR markers which was useful for further investigation.

Molecular techniques (RAPD and i-PBS) were used to study genetic diversity within persimmon collection at Horticulture Faculty of Mendel University in Lednice. The aim of the work was to distinguish 14 known and 6 of unknown origin persimmon cultivars. The basic screening of 20 OPT primers was applied to 4 cultivars differring in the place of origin. Within the group of screened primers there were chosen those, which gave polymorphic repeatable strong and middle strong bands. Selected primers were used for the RAPD reactions within the whole persimmon collection. Three OPA primers previously described in the literature were also used for the RAPD reactions within the whole persimmon collection. Additional 16 i-PBS primers previously described in the literature were also used for i-PBS analysis of the whole group of cultivars. Amplification was successful with 12 i-PBS primers. The FreeTree software package was used to gen erate a similarity matrix and then to produce a dendrogram using UPGMA analyses. The similarity dendrograms of all persimmon cultivars were created based on both approaches and also on combination of both analyses by program Tree View. All the dendrograms clearly separated the assessed cultivars into 4 clusters. There are cluster of American persimmons - Meader' (1), 'Garretson' (2) and 'Early Golden' (3). They are representatives of D. virginiana. Further part of dendrogram includes single D. lotus (5), which is also clearly separated from other cultivars of the genus Diospyros. The third cluster includes interspecific hybrids 'Rossiyanka' (10) and 'Nikitskaiya Bordovaiya' (13), which arised from crosses of D. virginiana and D. kaki. The last cluster is formed by cultivars of Japanese persimmon - 'Mikatani Gosho', 'Zenjimaru', 'Tone Wase', 'Hiratanenashi', 'Fuyu', Chinese cultivar - 'Sansi' and two Italian cultivars 'Vaniglia' and 'Tipo'. They are clustered without significant distinction. The similarities and the differences revealed among incorporation of cultivars into groups were compared with the literature findings.

ABSTRACT Persimmon is a deciduous and perennial tree of the Ebenaceae family. Diospyros kaki Thunb. and Diospyros lotus L. are the most common persimmon species in Turkey and cultivated especially in the northeastern parts of the country. This study was carried out to determine the mite species on D. kaki and D. lotus in the Ordu province (Black Sea region, Turkey) during 2012 and 2013. The survey was conducted weekly between April and October of each year. Leaf samples were collected from nine different districts (Altınordu, Çaybaşı, Fatsa, Gülyalı, İkizce, Kabadüz, Perşembe, Ulubey, and Ünye) of the Ordu province. Thirty-five mite species representing three orders were found in the surveys. Tydeus goetzi Schruft, 1972 (Prostigmata: Tydeidae) is newly discovered from Turkey and here described and illustrated. Brachytydeus paraobliqua (Panou & Emmanouel, 1996) (Prostigmata: Tydeidae) is re-described and its trito- and deutonymph are described for the first time. Four molecular markers, including inter-retrotransposon amplified polymorphism (IRAP), retrotransposon-microsatellite amplified polymorphism (REMAP), sequence-specific amplified polymorphism (SSAP), and amplified fragment length polymorphism (AFLP), were compared in terms of their informativeness and efficiency for analysis of genetic relationships among 28 genotypes in the genus Diospyros. The results were as follows: (1) the highest level of detected polymorphism were observed for IRAP; (2) AFLP was the most efficient marker system due to the simultaneous detection of abundant polymorphism markers per single reaction; (3) the marker index (MI) value was lower for SSAP than for AFLP, but SSAP had a higher level of detected polymorphism than AFLP; (4) the correlation coefficients of similarity were statistically significant for all four marker systems; (5) the four molecular markers yielded similar phylogenetic trees. To our knowledge, this was the first detailed report of a comparison of performance among three retrotransposon-based molecular markers (IRAP, REMAP, SSAP) and the AFLP technique (DNA-based molecular marker) on a set of samples of Diospyros. The results provide guidance for future efficient use of these molecular methods in the genetic analysis of Diospyros.
